Not the brochure version. The actual block-by-block reality of the role on a typical Tuesday.
Check overnight cultures — inspect iPSC colony morphology under the microscope, verify CO2 incubator temperature logs, and confirm media change was performed on schedule for HSC expansion flasks
Passage iPSC colonies or harvest HSCs for the day's flow cytometry panel — add ROCK inhibitor, detach colonies with ReLeSR, replate at optimal density, and document every intervention in Benchling (electronic lab notebook)
Set up and run flow cytometry — stain cells with a lineage marker panel (CD34/CD38/CD90 for HSC phenotyping; OCT4/NANOG for pluripotency), acquire on the lab's BD FACSAria, and export data files for FlowJo analysis
Lab meeting or journal club — present latest scRNA-seq UMAP data on iPSC-derived haematopoietic progenitors; receive questions from PI and lab members on whether the novel CD34+CD43lo cluster is a genuine progenitor or a cell-culture artefact
Lunch break and informal discussion with NCBS collaborators on campus — compare notes on a new organoid protocol published in Nature Methods and assess whether the CHIR99021 concentration needs adjustment for the current differentiation project
Bioinformatics analysis session — run Seurat pipeline on the institute's HPC cluster to re-cluster overnight 10x Genomics data, generate UMAP plots, and cross-reference marker gene lists with published bone marrow stem cell atlases
Write or revise paper section — draft the Results paragraph for the latest iPSC-HSC differentiation efficiency data, incorporate FlowJo export graphs, and respond to reviewer comment 3 from the pending Stem Cell Reports submission
Harvest cells for tomorrow's CRISPR electroporation — count cells on the Countess, centrifuge, resuspend in SE nucleofection buffer; check the gRNA efficiency T7E1 result loaded an hour ago on the gel imager; update DBT grant progress log
The real entry pathway for this role — eligibility, the qualifying exam, training, and licensing — in the order most people follow it.
B.Sc (Hons) in Biology, Biochemistry, Biotechnology, or Zoology from a tier-1 college (Delhi University, BHU, JNCU, Mumbai University, Christ University Bengaluru) followed by M.Sc. Alternatively, B.Tech Biotechnology from IIT/NIT/VIT for an industry-facing entry.
Fellowship gateway (mandatory for funded PhD): CSIR-UGC NET JRF (Life Sciences), DBT-BET JRF, or ICMR-JRF. DST-INSPIRE fellowship is available for top M.Sc ranks. Without a national fellowship, PhD seats at inStem, NCCS, CCMB, IISc, IIT Bombay, or AIIMS are not practically accessible.
The core credential. Choose labs with a clear stem-cell focus — inStem (iPSC, organoids, neural regeneration), NCCS Pune (haematopoietic stem cells, cancer stem cells), CCMB Hyderabad (developmental biology), AIIMS (translational HSC/MSC therapy), or IIT Bombay (tissue engineering and biomaterials). Supervisor and institute funding track record matter more than the institution name alone.
Post-doctoral training (typically required for PI track): 2-4 years at an international lab (Weissman lab Stanford, Srivastava lab UCSF/Gladstone, Gage lab Salk, or EU EMBL/Max Planck) OR at inStem/NCBS under a DBT-Wellcome India Alliance or Ramalingaswami Fellowship. Post-doc abroad dramatically expands iPSC, organoid, single-cell, and CRISPR skill sets unavailable in most Indian labs.
Syngene, Biocon, Stempeutics, and Hema Cord hire M.Sc graduates for process development, cell banking, and QC roles. PhD holders enter at scientist-I level. ICMR and DBT-BIRAC fund translational cell therapy projects through BIRAC CytoGenX and similar schemes.
The ICMR National Guidelines for Stem Cell Research (2021) and the Central Drugs Standard Control Organisation (CDSCO) cell therapy guidelines under the New Drugs and Clinical Trials Rules 2019 govern all clinical-grade work. Scientists on translational teams must understand IND filing, GMP cell manufacturing, and Institutional Committee for Stem Cell Research (IC-SCR) approval workflows.
Core skills you must own, the support skills you'll grow into, and the tools you'll have open all day.

DBT-inStem Community Portal / inStem Seminar Series
Institute mailing list / in-personinStem's public seminar series and researcher community hub in Bengaluru; all inStem/NCBS seminars are open to external researchers; the institute's mailing lists and Slack workspaces (accessible via institute affiliation) are the primary informal community for stem cell researchers in South India
r/stemcells
RedditEnglish-language subreddit for stem cell researchers and students globally; active discussion on protocol troubleshooting (iPSC culture failures, differentiation optimisation), career advice (post-doc applications, fellowship strategy), and new publications; useful for Indian researchers seeking international peer input on technical problems
ResearchGate Stem Cell Biology Network
ResearchGateThe stem cell biology topic group on ResearchGate where researchers share preprints, ask protocol questions, and find collaborators; heavily used by Indian researchers for literature discovery and for identifying international collaborators without institutional access barriers
The traps real practitioners wish someone had named for them in year one. Read these before you commit, not after.
Choosing a PhD supervisor based on institute prestige rather than lab publication track record
Treating IC-SCR approval as a bureaucratic formality rather than a regulatory foundation
Neglecting single-cell bioinformatics skills because 'someone else in the lab does it'
Applying for only one national fellowship (CSIR-NET or DBT-BET) without a backup plan
The upside that makes this work worth it, set honestly against the parts people quietly resent. Both sides, before you commit.
Straight answers to what people genuinely wonder before stepping into this work — no brochure spin.
Books, longreads, and references practitioners come back to.
