Not the brochure version. The actual block-by-block reality of the role on a typical Tuesday.
Arrive at THSTI; check overnight cell cultures (PBMC, DC, macrophage activation assays); record incubator CO₂ and temperature logs; review any failed wells from last night's ICS setup
Weekly lab meeting: present updated flow cytometry data on Th17/Treg balance in mucosal vaccination model; two postdocs present data from innate-cell TLR-agonist adjuvant experiments
PBMC processing — Ficoll gradient isolation from fresh blood samples collected from clinical study participants at the AIIMS collaborating clinic; aliquot and freeze for ELISPOT batches
Flow cytometry run: 10-colour panel for vaccine-induced B-cell subset characterisation (plasmablasts, memory B cells, germinal-centre phenotype markers); acquire on BD FACSCanto II; export FCS files to FlowJo
Lunch in institute canteen; check email — respond to reviewer comments on a paper under revision at npj Vaccines; check BIRAC INNOVATE portal for grant milestone document upload deadline
Data analysis: FlowJo gating on this morning's run; export MFI and frequency statistics to GraphPad Prism; build figure for group leader review next day
Journal club: one-hour discussion of a recent Cell paper on mRNA vaccine TFH cell priming in non-human primates — calibrate assay design against global standards
Write materials-and-methods section of the grant renewal (DST-SERB CRG scheme) — document the validated ELISPOT and ICS SOP, animal immunisation protocol, and IRB approval number for clinical sample collection
Supervise junior research fellow's first Western blot for checking antigen expression after lentiviral transduction; walk through blocking step and exposure time
Read two recent immunology papers; plan tomorrow's ELISA plate layout for cytokine multiplex; leave institute
The real entry pathway for this role — eligibility, the qualifying exam, training, and licensing — in the order most people follow it.
Route 1 — Research / Academic (dominant in India): BSc in Life Sciences, Microbiology, Zoology, or Biotechnology, followed by MSc in Immunology or Biochemistry, then PhD via CSIR-NET JRF, DBT-JRF, or ICMR-JRF. Top PhD destinations: NII Delhi (National Institute of Immunology), THSTI Faridabad (Translational Health Science and Technology Institute), NCBS Bangalore, IISc Bangalore (MRDG / MCNS), IISER Pune, CSIR-CDRI Lucknow, AIIMS Delhi. Fellowship stipends: CSIR/DBT JRF ₹37,000/month; SRF ₹42,000/month.
Route 2 — Vaccine / Pharma Industry: MSc Immunology, Microbiology, or Biotechnology from a DBT-recognised institution. Major hirers: Serum Institute (Pune), Bharat Biotech (Hyderabad), Biological E. (Hyderabad), Indian Immunologicals (Hyderabad), Biocon (Bangalore). Entry roles: bioassay scientist, immunogenicity testing, vaccine quality control (potency assays, ELISA, FACS-based immunoassays). PhD is valued but not always mandatory for junior scientist roles.
MBBS (NEET-UG) + MD Internal Medicine or Paediatrics (NEET-PG) + DM Clinical Immunology / DNB Allergy-Immunology (super-specialty). DM Clinical Immunology seats are available at AIIMS Delhi, PGIMER Chandigarh, SGPGI Lucknow, and JIPMER Puducherry. Rare but growing as autoimmune disease burden rises and biologics (TNF inhibitors, IL-6 blockers, JAK inhibitors) require specialist prescribers.
CSIR-NET (Life Sciences — Paper B covers immunology extensively); DBT-JRF (separate entrance for DBT-funded institutes); ICMR-JRF (for ICMR / NII / NIMR-track research); GATE-BT (for process-industry and vaccine-manufacturing roles requiring engineering exposure).
2-4 years standard for academic faculty track; recommended at NCBS, THSTI, NII, or international labs (NIH-funded, EMBL, Karolinska) for IISc/IIT/NII faculty aspirants. DBT Ramalingaswami Re-entry Fellowship and Wellcome DBT India Alliance Early Career Fellowship are the principal independent PI entry mechanisms in India.
Core skills you must own, the support skills you'll grow into, and the tools you'll have open all day.

The traps real practitioners wish someone had named for them in year one. Read these before you commit, not after.
Doing a PhD in immunology without clearing CSIR-NET or DBT-JRF first
Treating flow cytometry as an 'instrument I can pick up on the job'
Applying to pharma industry jobs with a purely academic immunology CV
Specialising in a research area with no Indian funding base
Ignoring bioinformatics and single-cell analysis skills
The upside that makes this work worth it, set honestly against the parts people quietly resent. Both sides, before you commit.
Straight answers to what people genuinely wonder before stepping into this work — no brochure spin.
Books, longreads, and references practitioners come back to.
